Anak dengan Down syndrome (DS) mengalami masalah keseimbangan tubuh yang dapat melemahkan daya tahan otot inti. Latihan melalui permainan, bagaimanapun dapat meningkatkan daya tahan otot inti. Oleh karena itu, riset ini bertujuan untuk menganalisis perbedaan pengaruh dan interaksi antara latihan berbasis permainan dan tingkat keseimbangan tubuh dinamis terhadap daya tahan otot inti anak dengan DS. Riset menggunakan sampel berjumlah 16 dibagi menjadi 4 kelompok berdasarkan tingkat keseimbangan tubuh dinamis (Tinggi vs. Rendah) dan latihan berbasis permainan (Ular Tangga vs. Engklek). Functional reach test sebagai tes keseimbangan dinamis dan daya tahan otot inti dinilai menggunakan plank test. Data selisih dari pretest dan posttest sebagai data yang dianalisis menggunakan teknik ANOVA dua arah dengan tingkat signifikansi 5%. Hasil riset menunjukkan efek utama yang signifikan untuk jenis latihan terhadap daya tahan otot inti, efek utama yang signifikan untuk tingkat keseimbangan terhadap daya tahan otot inti, dan interaksi yang signifikan. Engklek tepat untuk anak DS dengan keseimbangan dinamis tinggi, sedangkan Ular Tangga tepat untuk anak DS dengan keseimbangan dinamis rendah untuk meningkatkan daya tahan otot inti. Temuan ini dapat menjadi acuan pengembangan program latihan berbasis permainan yang disesuaikan dengan kondisi motorik anak-anak DS.
